This webinar examines scalable suspension culture of PSC-derived organoids using PBS Biotech Vertical-Wheel bioreactors. PBS Applications Engineer, Hannah Worden, explains how low-shear, homogeneous mixing and consistent geometry support organoid process development and scale-up. Case studies cover liver, pancreatic islet, and cerebellar organoids, including aggregate-size control, differentiation, functional markers, and improved organization under dynamic culture. With the audience, we address agitation, seeding density, media formulation, perfusion-based medium exchange, sampling, and…
This poster, presented at ISSCR Kyoto 2026, demonstrates a scalable strategy for iPSC manufacturing using Vertical-Wheel Bioreactors, addressing expansion, process development, cell banking, and differentiation. Linear scale-up from PBS-0.1 through PBS-80 produced 200 billion iPSCs at a 50 L working volume while maintaining pluripotency and normal karyotype. High-density work cell banks were developed through 3D adaptation and retained differentiation potential toward cardiomyocytes and pancreatic progenitors. MiniPRO further served as a controlled scale-down model, demonstrating growth…
This presentation, presented at ISSCR Kyoto 2026 explores how Vertical-Wheel Bioreactors can reduce development and manufacturing risk for PSC therapies by providing a scalable platform from process development through commercial production. It highlights global clinical trials supported by Vertical-Wheel technology, predictive scale-down with MiniPRO, controlled hydrodynamics, automation, and linear scale-up through PBS-80. Data demonstrate consistent PSC growth across scales, culminating in greater than 200 billion cells from a 50 L batch while maintaining pluripotency, illustrating a…
This guide collects and summarizes process parameters from successful experiments performed in Vertical-Wheel® bioreactors, across a variety of cell types including pluripotent stem cells, mesenchymal stem/stromal cells, exosomes and extracellular vesicles, organoids, and immune cells. Find the publication or resource most relevant to your own experiment to help with your process development.
This protocol provides detailed instructions for expanding mesenchymal stem cells on microcarriers using PBS-Mini bioreactors with Vertical-Wheel technology. The document covers the complete workflow from recovery of cryopreserved MSCs and expansion in adherent culture through inoculation, suspension culture, sampling, medium exchange, and harvest in three-dimensional bioreactor systems the PBS-Mini bioreactor. Recommended process parameters are based on internal optimization studies using multiple MSC lines and media formulations. Guidance is provided for microcarrier preparation, cell…
This protocol provides step-by-step guidance for forming and expanding pluripotent stem cell aggregates in PBS-MINI Vertical-Wheel® bioreactors. The workflow begins with thawing and adherent seed-train culture in T-flasks, followed by transfer to three-dimensional suspension culture in 0.1 L or 0.5 L vessels. Instructions cover inoculation, medium exchange, sampling, morphology assessment, metabolite and pH analysis, cell counting, viability testing, and harvest in the PBS-Mini bioreactor. The document presents recommended starting parameters for seeding density, working volume, agitation…
This publication reports a fully suspended process for expansion and six-stage differentiation of human induced pluripotent stem cells into stem cell-derived islets in 0.1 L and 0.5 L Vertical-Wheel® Bioreactors. A fivefold increase in vessel volume produced a twelvefold increase in islet-equivalent yield without compromising islet structure. The 27-day process generated endocrine populations expressing ?-, ?-, and ?-cell markers with glucose-responsive insulin secretion and transcriptomic characteristics approaching adult islets. The study also evaluated donor-derived iPSC lines, cellular…
This publication evaluates the robustness of a baseline process for serial expansion of human induced pluripotent stem cell aggregates in Vertical-Wheel bioreactors. Five commercial media and multiple cell lines were examined through short-term culture and serial passaging. Process outputs included growth kinetics, aggregate morphology, harvest efficiency, pluripotency, genomic stability, and response to stresses associated with in-vessel dissociation. All tested media supported spherical aggregate formation and strong expansion during an initial six-day culture, but important performance…
This brochure introduces PBS MiniPRO as an automated, small-scale Vertical-Wheel® Bioreactor system for cell therapy process development. The platform provides dissolved oxygen and pH control, closed-system operation, perfusion-capable workflows, and parallel operation. As a scale-down model of PBS manufacturing-scale Bioreactors, MiniPRO supports predictive process development and technology transfer. The brochure highlights improved process control, reduced variability, streamlined scale-up, and support for workflows requiring consistent aggregate formation and gentle mixing conditions.
This poster reviews key process development considerations required to achieve robust, efficient, and scalable manufacturing of allogeneic cell therapies**.** The authors discuss major scale-up challenges including equipment selection, hydrodynamic control, oxygen transfer, medium exchange, and cell harvest operations. Case studies demonstrate how process optimization and Vertical-Wheel bioreactor technology can reduce seed train requirements, shorten manufacturing timelines, decrease facility footprint, and integrate closed-system operation. The work also presents methodologies for…
This poster evaluates the PBS MiniPRO as a controlled scale-down platform for human induced pluripotent stem cell process development. The system combines four 500 mL Vertical-Wheel vessels with online pH and dissolved oxygen monitoring, gas control, integrated retention filters, perfusion pumps, and closed fluid-handling connections. Experiments examined process reproducibility, direct thaw from high-density cell banks, seed train, commercial and modified media, settlement and perfusion feeding, oxygen control, aggregate morphology, and sampling methods. Controlling dissolved oxygen at 50%…
This brochure introduces the PBS-0.5 Vertical-Wheel® PSC Perfusion Vessel, an integrated perfusion solution for pluripotent stem cell aggregate cultures and other large-particle processes. Continuous medium exchange through an integrated retention filter can support aggregate homogeneity, reduce manual handling, and enable downstream differentiation workflows. Comparative data show tighter aggregate-size distributions and smaller average aggregate diameters with perfusion than settlement-based medium exchange. The brochure also covers scalability, equipment compatibility, ordering…
